“Endgame” by Samuel Beckett Essay Example for Free

â€Å"Endgame† by Samuel Beckett Essay â€Å"Finished, it’s finished, nearly finished, it must be nearly finished Grain upon grain, one by one, and one day, suddenly, theres a heap, a little heap, the impossible heap†. (p. 3) Here the endgame starts. In chess the endgame is defined as the stage of the game when only few pieces are left on the board. The same is with the Beckett’s play as there are only four people left. The endgame is a series of moves when the game is near the ending and its outcome is decided before the ceremony of the endgame occurs. Samuel Beckett is known to be a chess player and in the play the author compares the endgame with the end of life – death as death is inevitable as the game always ends. We see that in the play the main characters are enacting repetitive rituals labeled as the part of their endgame. Despite inevitable outcome the main characters are dragging through the final moves making their routines and they are doing whatever it takes to get through the day and to see the next day. The characters try to get through despite their game is lost. The idea of chess game is put in accordance with movements on the stage. The most vulnerable piece on the board is Hamm who utters: â€Å"Me to play†. (p. 18) Hamm is perceived as the King and he seems to be the most powerful. Clov is protective character in the centre of the board and Hamm often relies on him. Therefore, Clov is presented as the Queen and he is able to move easily. However, his erratic way of movement seems to be more suitable to the movement of the Knight. Nell and Nagg are insignificant characters and they are viewed as the Pawns – they appear only when the King calls for them. One by one the main characters are dying and one by one the chess figures are leaving the board. Nothing is eternal in the world and every game has its ending. The theme of chess play shows cyclical Universe – ‘the play ends with a stalemate’. The game will be played over and over again. Works Cited Adorno, Theodor W. A Case Study Audit Report of Veterans Affairs Association

A Case Study Audit Report of Veterans Affairs Association Introduction The Veterans Affairs (VA) is subject to the Government Security Policy (GSP) and must ensure compliance with the GSP and operational standards. The VA is responsible for the conduct of an audit to determine the efficiency and effectiveness of its security program. At the request of the VA, we conducted an audit of security to provide management of the VA with an objective assessment of it security program. Overall, we found that the VA met the requirements of the Government Security Policy (GSP) with respect to compliance, efficiency, and effectiveness. The audit provides an overview of the main security measures we observed. We also identified areas for improvement. The department of Veterans Affairs Investigation A Case Study Audit Report Generally, the VA has put in place a security program which complies with the GSP and operational standards. The roles and responsibilities of Security Management, Personnel Security, Physical Security, Information Technology Security as well as Contracting Management Security and Contingency Measures Security are clearly defined in the Security Management Structure. The Departmental security officer (DSO) carries out his duties by coordinating, controlling and updating the security program on a regular basis. The VA has implemented adequate mechanisms to ensure the protection of sensitive information and assets. The sensitive information and assets are classified, designated, declassified or disposed of, in compliance with the standards. Emergency and recovery plans are periodically developed, documented and revised, in compliance with the requirements. Public Works and Secure Impact (PWSI) is currently responsible for security screening services which are conducted in compliance with the Security Policy and the Personnel Security Standards. Even though the original agreement between the two parties for this service is no longer valid. Moreover, certain roles and responsibilities between the two parties are not clearly established and defined in the agreement. Presently, the VA determines the security level related to the position requirements and requests the appropriate personnel screening. The PWSI acts as the administrative security officer by granting the level of security requested by the VA. About the Audit The Veterans Affairs (VA) is responsible for protecting sensitive data such as financial, medical, and personal Veteran and employee information under their authority. The information must be classified and designated considering the provisions for adequate exceptions of the Access to Information Act and the Privacy Act. The data appropriate to information technologies must be classified and specifically designated per their confidentiality, integrity, availability and value. Information and sensitive data must be protected per minimal standards, and related risk and threat assessment. The VA is responsible for the implementation of the Security Policy within its institution and must conduct an internal audit on their compliance with the policy and their efficiency in implementing it at least every year. This audit is conducted within the framework of Treasury Board Secretariats requirements in this respect. Objectives The objectives of the audit are to ensure the compliance of all sensitive information and goods with the Government Security Policy (GSP) and with the operational standards and the efficiency and effectiveness of the Security Program of the VA. More specifically, the objectives focused on: Security organization, Security Management, Physical Security and Personnel Security. Scope of the Audit The audit covers the following: Security Organization: the structure of security management at the VA for the overall security program. Security Management: the security program, the security education and training programs, the classification and designation of sensitive data, the measures of protection for sensitive information, the breaches and violations of security and other security-related incidents, the protection measures taken for external communications. Physical Security: the location and layout of installations, the identification and the application of protection measures in the installations, the examination and control of physical security measures. Personnel Security: the personnel security investigations, the authorization, refusal and revocation of security levels, the measures required at employees termination of employment. Security and management of emergency cases: necessary actions are taken to protect sensitive information and assets and employees during all types of emergencies. Security and management of contracting: security measurements are included with other requirements in contracts involving access to sensitive information. Approach and Methodology The audit methodologies are comprised of interviews, data gathering, information and report analyses, the study of files and the observation of practices. Findings and Management Responses Security Organization Objective: To verify whether there is in place a security management structure meeting the Agencys requirements for the overall security program, specifically management security, physical security and personnel security. VA has implemented a security management structure which meets the overall security program needs of the Agency. The security responsibilities are clearly defined, established and assigned to personnel whose positions include security responsibilities defined in the position description. Secure Impact, a tenant in the same building as VA, is responsible for the development and implementation of the physical security. For personnel security screening VA depends on the services of PW. Area of Improvement The audit has found that the agreement between the VA and PW for the delivery of personnel security screening services has expired. Furthermore, certain roles and responsibilities of PW as related to the security of the VA personnel were not clearly established in the expired agreement. Management Response The VA recognizes the importance of maintaining valid agreements with its service providers, especially when dealing with security issues. The VA also appreciates the necessity of having clear roles and responsibilities defined in the agreement and understood by all parties. After being apprised of the above situation, the VA contacted PW to begin negotiation on a new agreement, which would clearly state roles and responsibilities of all parties. The VA will also ensure that this agreement is revised periodically and that it is extended, based on operational requirements. Security Management Objective: To verify whether a good security program is an integral part of the VAs overall program and meets the GSP requirements and operational standards. The VA currently has a good security program in place which complies with the requirements of the GSP and operating standards. The responsibilities assigned to security personnel are fully carried out. Guides and procedures have been developed which are used as guidelines for those in charge of security. Area of improvement Develop a security policy or adapt the TBS security policy to meet the VA requirements. Management Response The VA will review current Government Security Policy and determine how and if it can be adapted to meet VA requirements. Should this not be feasible, the VA will develop its own internal security policy. It should be noted that although the VA has no official internal policy which covers all aspects of security, it does have a policy on electronic mail, which sets out standards for ensuring that established security levels are adhered to and that needed information is preserved. Objective: To verify whether there are good security education and training programs. The VA does not have in place a security education and training program. Area of improvement Provide training to employee with security responsibilities. Management Response The VA is fully supportive in providing training to its employees. Each year, a training plan is submitted by employees and approved by the Chairperson. The VA will ensure that those employees with specific security functions are made aware of and encouraged to take training necessary to meet current and upcoming security requirements. Objective: To verify whether sensitive information is classified and designated in compliance with the GSP and operational standards, and whether the classifications and designations are unclassified or eliminated when the information is no longer, or less of a sensitive nature. The VA has implemented a mechanism to ensure that goods of a sensitive nature are classified and designated in compliance with the GSP and operational standards; the same mechanism is also being used to declassify or dispose of the same goods. Area of improvement No recommended improvement Objective: To verify whether protection measures are applied for sensitive information, as well as for employees, in compliance with the mandatory standards and with a risk management methodology. The VA has implemented mechanisms to ensure the security of sensitive information. A process is in place to declassify sensitive information when it is no longer sensitive. The controls in place ensure authorized to receive such information. Area of improvement No recommended improvement Objective: To verify whether breaches of security, security violations and other security-related incidents that may happen are the subject of an enquiry, that measures are taken to minimize the losses and that the necessary administrative or disciplinary measures are taken if warranted. Breaches of security, security violations and other security-related incidents are reported to Secure Impact. Secure Impact is responsible to take the necessary administrative measures and to ensure follow-up. A mechanism is in place and is used to report security breaches and to prepare reports. Area of improvement No recommended improvement Objective: To verify whether the necessary protection measures are taken for the sensitive information communicated to or from official sources outside the department. The VA follows procedures concerning sensitive information transmitted to official sources outside the department. Area of improvement No recommended improvement Physical Security Objective: To verify whether consideration was given to providing good siting to, as well as adequate retrofit of installations, to reduce or eliminate threats and risks to which the information, and the employees in those installations are exposed. The VA uses the facilities along with other government departments. Secure Impact ensures the physical security, thus reducing or eliminating threats and risks. A physical security committee is established with a representative of the VA. In this regards, the physical security is adequate. Area of improvement No recommended improvement Objective: To verify whether the required physical protection measures are applied in installations, so that sensitive information is well protected. The current physical protection measures ensure that sensitive information is protected. Area of improvement No recommended improvement Objective: To verify whether the physical security measures required are applied in the installations to ensure the protection and security of staff. Implemented physical security measures in the VA facilities ensure employee protection and security. Area of improvement No recommended improvement Objective: To verify whether the physical security measures are periodically reviewed and controlled. Security measures are reviewed and controlled periodically. Area of improvement No recommended improvement Personnel Security Objective: To ensure that the personnel of the VA is subjected to a security check per the Government Security Policy (GSP) and the standard on Personnel Security The audit found that security checks were conducted in compliance with the Government Security Policy (GSP) and the standards on Personnel Security. PW is responsible for the safe storing of personnel records and for the filling in and storing of security investigation forms requests. Area of improvement No recommended improvement Objective: To verify whether the necessary levels of security are authorized, refused and revoked per the GSP and to the personnel security standard, and whether such measures are taken in a just and impartial way. The VA has no record of refusals or revocations of levels of security. The VA recognizes its responsibilities in this matter. Area of improvement No recommended improvement Objective: To verify that the necessary measures are taken to reduce or eliminate any risk for the sensitive information and goods as well as for the departments essential systems at the termination of employment. The audit found that the necessary measures are taken at the termination of employment. Area of improvement No recommended improvement Security and Contracting Management Objective: Ensure that security requirements are included with other requirements in contracts when they involve access to sensitive information. The VA does not have mechanisms in place to check authorization to access facilities by the contracting parties. Area of Improvement Put in place a mechanism to check the authority to access the facilities by the contracting parties. Management response The VA is fully aware of its responsibility to ensure that only those individuals with proper authority are given access to its facilities. In some cases, authority to access VA facilities is given by another department, such as Secure Impact, but the VA is informed in advance. The VA will ensure that in those situations where another department gives access to its facilities, once the individuals show up, their name and authority will be verified with the other department. Conclusion The audit provides an overview of the main security measures observed, as well as, identifies areas for improvement. The audit methodologies are comprised of interviews, data gathering, information and report analyses, the study of files and the observation of practices. Finally, the audit covers security organization, security management, physical security, personnel security, security and management of emergency cases, and security and management of contracting. Subcortex Diencephalon lesions from the view of the Neurologist and the Neurosurgeon :: Medical Medicine Essays

Subcortex Diencephalon lesions from the view of the Neurologist and the Neurosurgeon The diencephalon, which includes the thalamus, hypothalamus, and subthalamus of the brain will be discussed in this project paper however, the main focus will by on the hypothalamus of the diencephalon structure. To begin a brief review of the functions of the thalamus and hypothalamus, the thalamus is the input center for sensory information and the output center for motor information. The hypothalamus regulates hunger, thirst, and the body’s temperature. The hypothalamus also releases hormones and plays a role in sexual responses and behaviors, and our circadian rhythms (Campbell, 1999). The hypothalamus can be affected in many ways from genetics/hormones to injury from a lesion or accident. If a female fetus is exposed to too much testosterone during their critical period the hypothalamus can become masculinized which would lean create masculine behaviors in the female. Those fetuses that are male but are androgen insensitive have femininized hypothalamuses and portray feminine behaviors (Kalat, 2004). Damage such as a lesion to the lateral hypothalamus can cause an individual to refuse food and/or water depending on where the lesion is located. The opposite of true of damage occurs in the medial hypothalamus, the individual will over eat or drink (Kalat, 2004). The Neurologist The neurologist can perform many experiments on the hypothalamus by placing electrodes on the different parts, which simulates what could happen to the individual if this portion of the hypothalamus became injured. Studies have shown that the hypothalamus control functions such as mating, urination and defecation, eating and drinking, temperature regulation (through homeostasis), and circadian rhythms (Farr, 2002). The anterior portion of the hypothalamus is involved in copulation however; the septal region is also involved. When electrical stimulation is directed to this area the individual will experiences sexual emotions and thoughts. A study was done on cats where electrical stimulation on the anterior portion of the hypothalamus was done thus showing that the cat acted as though it was going to urinate or defecate along with the other processes such as hiding/covering the feces (Farr, 2002). One of the most important functions of the hypothalamus is in the lateral and ventromedial regions, these areas in the hypothalamus control eating and drinking. If a lesion in the lateral region of the hypothalamus is created experimentally or damage is done to

The American Economy Essay -- Economics Governmental Automobiles Essay

The American Economy The American economy is a vibrant, free-market system that is constantly developing out of the choices and decisions made by millions of citizens who play multiple, often overlapping roles as consumers, producers, investors and voters. The changes in the organization and performances of the manufacturing industry over the last century have helped shape the American economy. The Automotive industry perhaps made the biggest changes to their manufacturing processes. I will be reviewing the role of the industrialist Henry Ford and his innovative methods that changed the organization and performance of the American manufacturing industry forever. He produced an affordable car, paid high wages and helped create a middle class, which in turn fueled the America Industrial revolution into overdrive mode. I will also review the impact of these performance and organizational changes on the service sector and the agricultural industry. But first we look at the automotive industry. The requirements for mass production of a particular commodity must include the existence of mass consumption of the commodity, sufficient to justify large investment. Most early automobile companies were small shops, hundreds of which each produced a few handmade cars, predominately sold to the rich. In America almost all of the producers were assemblers who put together components and parts that were manufactured by separate firms. The assembly technique also lent itself to an advantageous method of financing. It was possible to begin building motor vehicles with minimal investment of capital by buying parts on credit and selling the finished cars for cash; the cash sale from manufacturer to dealer has been integral in the marketing of motor vehicles in the United States ever since. The outstanding contribution of the automotive industry to technological advance was the introduction of full-scale mass production, a process combining precision, consistency, interchangeability, organization, and continuity. America, with its large population, high standard of living, and long distances, was the natural birthplace of the technique, which had been partly explored by agricultural methods, in the 19th century. The U.S. role was emphasized in the popular description of standardization and interchangeability as "the American system of manufacture." The fundamental te... ... But during times of hardship, economy downturns or even long-term recession, a basic instinct of survival takes over. As a rule, consumers look for the best values for what they spend, while producers seek the best price and profit for what they have to sell. Government, at the federal, state and local levels, seeks to promote the public safety, assure reasonable competition. The U.S. economy has changed in other ways as well. The population and the labor force have moved dramatically from farms to cities, from fields to factories and, above all, to service industries. In today's economy, the providers of personal and public services far outnumber producers of agricultural and manufacture goods. The consumer will exert a measure of influence over the market economy. Naturally, most consumers look for good values when they buy, as well as product reliability and safety. If one automobile manufacturer, domestic or foreign, produces a better car at a lower price, the market will b egin to shift as that car attracts more sales than its competitors. In theory, this phenomenon rewards efficient producers who maintain high quality at low prices, and drives out those who cannot compete.

The Fall of the House of Usher :: Essays Papers

The Fall of the House of Usher The story starts out with the narrator riding up to an old and gloomy house. He stresses that the overall persona of the house is very eerie. The reason he is at this house is because he received a letter from an old friend by the name of Roderick Usher. Roderick and the narrator were intimate friend at a young age but they had not spoken to each other in several years. The narrator examined the house for a great time as he rode toward the house, he noticed that the house had been severely neglected over time. That the house’s beautiful woodwork and Gothic type of architecture have not been maintenance to any degree since he had last seen it. When he arrived at the home the servant who took his hoarse and directed him to the room that Mr. Usher was in greeted him. Inside the house was also very ornate, but it to had also been left alone for to long. The entire house had a gloomy atmosphere that would put a chill down most people’s spines. When he entered the room his friend was staying in he was warmly welcomed. He could not believe the changes that his dear childhood friend had endured. Roderick was very ill for he was not able to do many things. He could only listen to certain types of sounds, eat certain foods, and he could only look at certain things. Together they to loved to read books of Gothic natures that only added to the ora of the house. Usher had a great collection of books but his favorite was the manual of the forgotten church. Roderick’s sister was also very ill with something that was never diagnosed. In the first few days of the narrator’s visit Ethelred died. She was put into a casket and then the two men carried her to a vault that was below the narrator’s room. Several night later the men noticed sound coming from the vault and went down to investigate. They began to read to each other again, but Roderick paid no attention.

Key Economic Factors of Microsoft Corporation Essay

As a dominator in both the PC operating system and office suite markets, Microsoft Corporation expose themselves to the cloud-based computing model presents execution and competitive risks. They are transitioning to a computing environment characterized by cloud-based services used with smart client devices. Their competitors are rapidly developing and deploying cloud-based services for consumers and business customers. The pricing and delivery models are evolving, and cause economic growth to move slower. They may not be able to adequately protect their intellectual property rights. Protecting their global intellectual property rights and combating unlicensed copying and use of software and other intellectual property is difficult. While piracy adversely affects U.S. revenue, the impact on revenue from outside the U.S. is more significant, particularly in countries where laws are less protective of intellectual property rights. As a result, their revenue in these markets likely will grow slower than the underlying PC market. Similarly, the absence of harmonized patent laws makes it more difficult to ensure consistent respect for patent rights. Throughout the world, they actively educate consumers about the benefits of licensing genuine products and obtaining indemnification benefits for intellectual property risks, and they educate lawmakers about the advantages of a business climate where intellectual property rights are protected. However, continued educational and enforcement efforts may fail to enhance revenue. Reductions in the legal protection for software intellectual property rights could adversely affect revenue. In conclusion, the economic growth is the main key factor for Microsoft Corporation and this reason likely to continue due to the globally affect issue on the intellectual property rights and unlicensed copying and use of software worldwide. The company in its life cycle Microsoft Corporation is at the stages of maturity, as they are in the market of computing for a few decades. This stage will remain as they are a dominator in the PC operating system and office suite markets. They are devoting significant resources to develop and deploy their own competing cloud-based software plus services strategies. While they have their own expertise, investments in infrastructure, and the breadth of their  cloud-based services provides them with a strong foundation to compete and to remain as a strong player in the market. The five or six most important factors for success in this business In order to success, there is a few factors in Microsoft Corporation including as follow: continuing to innovate and bring to market compelling cloud-based experiences that generate increasing traffic and market share; maintaining the utility, compatibility, and performance of our cloud-based services on the growing array of computing devices, including smartphones, handheld computers, netbooks, tablets, and television set top devices; and continuing to enhance the attractiveness of our cloud platforms to third-party developers. continuing to make significant investments in research, development, and marketing for new products, services, and technologies, including the Windows PC operating system, the Microsoft Office system, Bing, Windows Phone, Windows Server, Zune, Windows Live, the Windows Azure Services platform and other cloud-based services offerings, and Xbox 360. To create competing vertically-integrated model, in which a single firm controls both the software and hardware elements of a product, has been successful with certain consumer products such as personal computers, mobile phones, and digital music players. We also offer vertically-integrated hardware and software products This company stand with respect to these factors? Microsoft Corporation will stand with respect to these factors with the help of talented employee. As their business depends on the ability to attract and retain talented employees. Their business is based on successfully attracting and retaining talented employees. The market for highly skilled workers and leaders in the industry is extremely competitive. They are limited in their ability to recruit internationally by restrictive domestic immigration laws. If they are less successful in their recruiting efforts, or if they are unable to retain key employees, their ability to develop and  deliver successful products and services may be adversely affected. Effective succession planning is also important to their long-term success. Failure to ensure effective transfer of knowledge and smooth transitions involving key employees could hinder their strategic planning and execution. Four key business risks related to client’s business and industry. They may not be able to adequately protect their intellectual property rights. Protecting their global intellectual property rights and combating unlicensed copying and use of software and other intellectual property is difficult. While piracy adversely affects U.S. revenue, the impact on revenue from outside the U.S. is more significant, particularly in countries where laws are less protective of intellectual property rights Third parties may claim we infringe their intellectual property rights. Due to the claiming, Microsoft has to stop selling or redesign affected products, or pay damages to satisfy indemnification commitments with their customers. They may not be able to protect their source code from copying if there is an unauthorized disclosure of source code. Department of biology Essay

INTRODUCTION: Every cells of living organism contains genetic materials known as deoxyribonucleic acid, DNA. It can be isolated from tissue sample of living things by separating it from other cellular component in a manner that still preserves its structures. The structure of DNA is double-stranded helices that made up from the monomer of nucleotides. Each of the nucleotides composed of three parts that are the phosphate group, deoxyribose sugar backbone and also nitrogenous bases (Adenine, Guanine, Thymine and Cytosine). This nitrogenous base are arranged in sequence and holds the information and coding for controlling the physical trait that we all have as well as the regulation of our body. DNA extraction is simply a process that results in separation of DNA from the cells or viruses that are hosting it. Through the meaning is simple but the process is not. When we go into the peculiar details of DNA extraction, we realize that it’s more of an initial stage in other intensive DNA testing processes. DNA test could be performed for any reason; however for any DNA testing to happen the first stage normally is the isolation and extraction of DNA molecules from the cells that they reside in. DNA extraction follows a series of step, stripping all proteins from the DNA and the extraction protocols have to make sure that the DNA thus obtained via isolation and extraction is of high or acceptable quality. DNA isolation is a process of purification of DNA from sample using a combination of physical and chemical methods. Currently it is a routine procedure in molecular biology or forensic analysis. Primary structure consists of linear sequence of nucleotides that are linked together by phospodiester bonds. It is the linear sequence of nucleotides that make up the primary structure of DNA. Specific techniques must be chosen for isolation of DNA from some samples such as samples from microorganisms with thick cellular wall, for example yeast. Biological DNA represents the information which directs the functions of a living thing. A. Yeast DNA Extraction Materials : 1 packet of dry yeast, Sodium chloride, Meat tenderizer, Ice cold 95% ethanol, Sunlight detergent, distilled water, blender, graduate cylinders ( 10mL, 100mL and 500mL ), Beaker ( 250mL, 100mL ), glass stirring rod and wooden sticks, 15mL test tube, test tube rack, 1 ,l pipette and blue tips. The test tube was tilted to a 45 degree position. 3 ml of 95% ice cold ethanol was gently added to the side of the tube. 10. The test tube was leave undisturbed for 3-5 minutes. A layer will be formed in the tube. 11. DNA precipitate was formed at the interphase layer. A wooden stick was used to swirl the DNA out. Result : Table C(i) : Orange DNA Extraction Table C (ii) : Apple DNA Extraction RESULT ALL OF EXPERIMENTS: Sample Extraction Amount Apple Success Large Onion Success Small Orange Success Small Yeast Success Small DISCUSSION: Based on our experiment discussion, we obtained that the difference amount of DNA extraction from yeast, onion, apple and orange is different. The amount of apple DNA extraction is larger than onion, orange and apple DNA extraction. Each step of the process will help in a certain way to extract DNA, until we are finally successful in the end. In the yeast, extraction, the ethanol will be able to separate the DNA and it will float between the less dense ethanol and the denser homogenizing mixture. In the onion DNA extraction, the chloroform and homogenizing medium will help break down the cell membranes and the ethanol like in the yeast DNA extraction. It will cause the DNA to separate and be suspended in the interface between the two solutions. It will be same to apple and orange DNA extraction. The precaution step on this experiments we tilt 45Â º to add the ethanol. This is because it will form a layer on top of the sample because the ethanol is less than water. So, it does will be on the top of layer. We also used 50ï‚ ° of hot water in yeast solution. Its will formed the best of result because hot water is the optimum temperature for yeast. We also obtained about perceptions step why only clear solution produced in these experiments. Its means the extraction was failed. CONCLUSION: REFERENCES: http://www.whatisdna.net/dna-extraction.html http://classic.sidwell.edu/us/science/vlb5/Labs/DNA_Extraction_Lab/dna_extraction_lab.html#predictions http://sciencehk.weebly.com/lab-reports.html ANSWER AND QUESTION: Answer all the questions. 1. Describe the functions of following: a. Applying blender to sample and salt/detergent solutions Strain the DNA mixture. b. Salt Salty water helps the DNA precipitate (solidify and appear) when alcohol is added. c. Detergent Detergents are used to break down cell walls and nuclear membranes to release the DNA. They work by chemically poking holes in the cell membranes or walls. Once holes are poked in the membranes, the membranes can be further distrupted mechanically, as with a blander. After that, it’s easier to get the contents of the cell out, including the DNA, d. Meat tenderizer solutions Meat tenderizer acts as an enzyme. The DNA in the nucleus of the cell is moulded, folded, and protected by proteins. The meat tenderizer cuts the proteins away from the DNA. e. 95% ice cold ethanol Having ice cold ethanol only increases the rate of precipitation of DNA and helps increase yield of DNA. It can either use room temp or ice cold ethanol for DNA precipitation. Think about precipitation of a super concentrated solution as you decrease the temperature. As the temperature decreases, the amount of precipitation increases. Overall temperature affects solubility. As temperature decreases the substance becomes more insoluble (in general. this does not apply to every molecule). So, ice cold EtOH allows for more DNA to interact together and allow for a more rapid and efficient precipitation of DNA. 2. Compare the reliability amount of DNA obtained from all the samples. The reliability amount of DNA obtained from apple is much larger compared to Reliability amount of DNA from onion, orange and apple. 3. Write out the principle involved in DNA extraction. – Break open cells by mashing the fruit. – Dissolve organelles and cell membranes with detergent. – Separate DNA from proteins with salt – Filter out the clumps with a coffee filter – DNA precipitates in cold alcohol and is spooled out.
